实验方案

样品实验方案 

溶液制备 

1.储备溶液

所有未使用的储备溶液应分为一次性使用的等分试样,并在制备后储存在-20°C下。 避免重复冻融循环。
酪蛋白,TAMRA共轭原液:
在PBS缓冲液中制成5-10 mg / mL酪蛋白,TAMRA偶联的原液。 注意:未使用的储备溶液可分为一次性使用的等分试样,并在-20 oC下保存,并避免暴露在光线下。

 

2.工作溶液

酪蛋白TAMRA共轭的工作溶液(2X):
将TAMRA共轭的原液以100-400μg/ mL的浓度稀释至50-100 mM Tris缓冲液(pH 7.4)中。 2X Assay工作溶液设计用于检测胰凝乳蛋白酶,胰蛋白酶,嗜热菌蛋白酶,蛋白酶K,蛋白酶XIV和人白细胞弹性蛋白酶的活性。 对于其他蛋白酶,请参考表1中合适的测定缓冲液配方。 对于每种蛋白酶,应通过实验确定测定工作溶液的最佳浓度。

 

样品操作及分析

1.将等体积的胰蛋白酶标准品或样品与2X Assay工作溶液混合。

2.在Ex / Em = 490/525 nm处监控荧光的增加。

2.1 对于动力学读数:立即开始连续连续测量荧光强度,并每5分钟记录一次数据,持续30分钟。

2.2 对于终点读数:在避光的条件下,将反应在所需温度下孵育30至60分钟。 然后测量荧光强度。
